Here we describe the molecular identification of a cDNA encoding a novel serine protease we have termed protease T. The deduced amino acid sequence encodes a prepro form of 290 amino acids, and its alignment with other well- characterized serine proteases indicates that it is a member of the S1 serine protease family. We have found that the protease T mRNA is expressed in stomach, testis, retina, fibroblasts, spinal cord, and several regions of the brain. Protease T mRNA is also found in leukocytes and in the Jurkat (ATCC TIB- 152) T cell line. Thus, this protease is potentially involved in gastric, testicular, retinal, dematological, neurological/neurodegenerative and/or immunological disorders. The protease T gene maps to human chromosome 16p13.3 which is near the tryptase locus. Enzymatically active protease T, we have generated, is amenable to further biochemical analyses for the identification of physiological substrates and specific modulators.

Cette invention décrit l'identification moléculaire d'un ADNc codant une nouvelle sérine protéase appelée protéase T. La séquence d'acides aminés déduite code une forme prépro de 290 acides aminés, et son alignement avec d'autres sérine protéases bien caractérisées indique qu'elle est un membre de la famille des sérine protéases S1. On a découvert que l'ARNm de la protéase T est exprimé dans l'estomac, les testicules, la rétine, les fibroblastes, lamoelle épinière et plusieurs régions du cerveau. On a également trouvé l'ARNm de la protéase T dans des leucocytes et dans la lignée des lymphocytes T de Jurkat (ATCC TIB-152). Cette protéase est donc potentiellement impliquée dans les troubles gastriques, testiculaires, rétiniens, dermatologiques, neurologiques/neurodégénératifs et/ou immunologiques. Le gène de la protéase T trouve sa correspondance avec le chromosome humain 16p13.3 qui est proche du locus de la tryptase. La protéase T à action enzymatique, qui est produite ici, est susceptible d'autres analyses biochimiques pour l'identification de substrats physiologiques et de modulateurs spécifiques.
