A method of expression DNA in a cell comprises, in a cell that expresses a replication factor, transfecting the cell with a vector, wherein (i) the vector contains a DNA, or is adapted to receive a DNA, in operative combination with a promoter for expression of the DNA; and (ii) extrachromosomal replication of the vector is dependent upon presence within the cell of the replication factor. Stable maintenance of the vector and expression of the DNA is thereby achieved. Vectors for expression of DNA are provided as are assays for the effect of expression of DNAs in cells, the DNAs being expressed from the vectors.
L'invention concerne un procédé d'expression de l'ADN dans une cellule. Ce procédé consiste à transfecter avec un vecteur une cellule exprimant un facteur de réplication, i) ce vecteur renfermant un ADN, ou pouvant recevoir un ADN, et étant associé de manière fonctionnelle à un promoteur afin d'exprimer cet ADN, et ii) la réplication extra-chromosomique dudit vecteur dépendant de la présence dudit facteur de réplication à l'intérieur de la cellule. On assure ainsi le maintien stable du vecteur et l'expression de l'ADN. L'invention concerne également des vecteurs d'expression de l'ADN, ainsi que des dosages de l'effet de l'expression des ADN dans les cellules, ces ADN étant exprimés à partir des vecteurs.
