The present invention relates to a method of detecting a mutator phenotype of tumor cells in a patient, which comprises the steps of: a) obtaining a genomic DNA sample of the tumor; b) obtaining a genomic DNA sample of a tumor-free tissue of the patient; c) subjecting the DNA samples of steps a) and b) to amplification using primers which are flanking repeat pattern characteristic of a mutator phenotype or non-radioactive inter-Alu PCR; d) subjecting the amplified fraqments of step c) to electrophoretic fractionation on a polyacrylamide gel to determine the presence of a variation in band profile between tumor and tumor-free DNA indicating a genomic instability associated with a mutator phenotype followed by hybridization with a probe corresponding to an instability prone locus; and e) comparing the allelic status of at least one instability prone locus in the DNA samples of step c).
La présente invention concerne une méthode pour détecter un phénotype mutateur des cellules tumorales chez un malade, qui comprend les étapes suivantes : a) obtenir un échantillon d'ADN génomique de la tumeur; b) obtenir un échantillon d'ADN génomique de tissu non tumoral du patient; c) soumettre les échantillons d'ADN provenant des étapes a) et b) à une amplification à l'aide d'amorces qui sont des séquences répétitives flanquantes caractéristiques d'un phénotype mutateur ou à l'inter-Alu PCR non radioactive; d) soumettre les fragments amplifiés de l'étape c) à un fractionnement par électrophorèse sur gel de polyacrylamide pour déterminer la présence d'une variation dans le profil de bandes entre l'ADN tumoral et l'ADN non tumoral indiquant une instabilité génomique associée à un phénotype mutateur, puis à une hybridation avec une sonde correspondant à un locus sujet à instabilité; et e) comparer l'état allélique d'au moins un locus sujet à instabilité dans les échantillons d'ADN de l'étape c).
