The invention provides a biograting for use in determining the amount of an analyte in a sample in a light diffraction assay. The biograting has a smooth, solid surface that reduces nonspecific binding, thereby providing better resolution than in prior art systems. A process for manufacturing the biograting comprises adhering a uniform layer of hybridizing reagent on the smooth, solid surface and exposing the surface to UV radiation through a shadow mask with diffraction grating lines to selectively deactivate the hybridizing reagent. This process produces a biological diffraction grating design of lines of active hybridizing reagent. The invention includes a biograting wherein the hybridizing reagent is a nucleotide sequence. This biograting is used to detect a specific DNA sequence in a diffraction hybridizing assay.
