The present invention relates to the use of a plasmid encoding Staphylococcus aureus polypeptides and its use in the preparation of compositions and vaccines. More specifically, the present invention is concerned with compositions, DNA vaccines and methods for providing an immune response and/or a protective immunity into mammals against a Staphylococcus aureus associated disease, such as mastitis. The plasmid used in the composition or DNA vaccine comprises at least one nucleotide coding sequence of a Staphylococcus aureus polypeptide, such as the Clumping factor A (CIfA), the fibronectin-binding protein A, the sortase-A or the pre-pheromone (ArgD).
