The present invention relates to targeted delivery systems for delivering therapeutic agents to tumor. The invention further relates to methods of delivering a therapeutic agent to a tumor for the prevention and treatment of cancer by killing tumor cells and tumor associated endothelial cells. In particular, the present invention provides a tumor-targeted drug delivery system comprising a NGR-containing molecule linked to a delivery vehicle encapsulating a therapeutic agent, preferably a drug, such as a cytotoxic agent or a chemotherapeutic agent. Specifically, the delivery systems of the present invention are capable of delivering an increased amount of therapeutic agent to a tumor as compared to other delivery systems. In particular, the delivery systems of the present invention are capable of accumulating a higher amount of therapeutic agent in a tumor, or in the vicinity of a tumor cell or tumor-supporting cell, resulting in exposure of the tumor cell and tumor associated endothelial cell to therapeutic levels of the agent for a longer period of time as compared to other delivery systems. The present invention also describes pharmaceutical compositions comprising the delivery systems of the present invention. The present invention further relates to a tumor treatment comprising an increased amount of therapeutic agent delivered by the system of the present invention as compared to other delivery systems. The delivery systems and pharmaceutical compositions can be administered to a subject, preferably a human, alone or in combination, sequentially or simultaneously, with other prophylactic or therapeutic agents and/or anti- cancer treatments.
L'invention concerne des systèmes de libération ciblée permettant de libérer des agents thérapeutiques sur une tumeur. L'invention concerne également des méthodes de libération d'agent thérapeutique sur une tumeur, utilisées dans la prévention et le traitement du cancer par l'élimination des cellules tumorales et des cellules endothéliales liées à une tumeur. L'invention concerne en particulier un système de libération de médicament ciblée sur la tumeur, comprenant une molécule contenant du NGR, liée à un excipient de libération encapsulant un agent thérapeutique, de préférence un médicament, tel qu'un agent cytotoxique ou un agent chimiothérapeutique. Les systèmes de libération selon l'invention présentent la spécificité de libérer une quantité accrue d'agent thérapeutique sur une tumeur par rapport aux autres systèmes de libération. Les systèmes selon l'invention peuvent en particulier accumuler une quantité supérieure d'agent thérapeutique dans une tumeur, ou à proximité d'une cellule tumorale ou d'une cellule soutenant une tumeur, ce qui permet d'exposer la cellule tumorale et la cellule endothéliale liée à une tumeur à des niveaux thérapeutiques d'agent, pendant une période plus longue que dans les autres systèmes de libération. La présente invention concerne également des compositions pharmaceutiques contenant lesdits systèmes de libération. L'invention concerne encore un traitement tumoral contenant une quantité accrue d'agent thérapeutique libéré par rapport aux autres systèmes. Les systèmes de libération et les compositions selon l'invention peuvent être administrés à un sujet, de préférence un humain, seul ou en combinaison, de manière séquentielle ou simultanée, avec d'autres agents prophylactiques ou thérapeutiques et/ou traitements anticancéreux.
