A transcutaneous immunization system delivers antigen to immune cells through the skin, and induces an immune response in an animal or human. For example, a skin-active adjuvant (e.g., an ADP-ribosylating exotoxin) can be used to induce an antigen-specific immune response (e.g., humoral and/or cellular effectors) after transcutaneous application of a dry formulation containing antigen and adjuvant to skin of the animal or human. The dry formulation may be a powder or a unit-dose patch. Use of adjuvant is not required if the antigen is sufficiently antigenic. Transcutaneous immunization may be induced with or without penetration enhancement.

L'invention concerne un système d'immunisation transcutanée permettant d'administrer un antigène aux cellules immunitaires à travers la peau, et d'induire une réponse immunitaire chez un animal ou un être humain. Le système utilise, par exemple, un adjuvant actif sur la peau (notamment, une exotoxine ribosylant l'ADP) pour induire une réponse immunitaire spécifique de l'antigène (effecteurs humoraux et/ou cellulaires par exemple) après application transcutanée, sur la peau de l'animal ou de l'être humain, d'une formulation sèche contenant un antigène et un adjuvant. Ladite formulation sèche peut être une poudre ou un patch monodose. L'utilisation de l'adjuvant n'est pas nécessaire si l'antigène est suffisamment antigénique. Il est possible d'induire l'immunisation transcutanée en ayant éventuellement recours à un agent facilitant la pénétration.
