Pregnancy-associated glycoproteins (PAGs) are structurally related to the pepsins, thought to be restricted to the hoofed (ungulate) mammals and characterized by being expressed specifically in the outer epithelial cell layer (chorion/trophectoderm) of the placenta. By cloning expressed genes from ovine and bovine placental cDNA libraries, the inventors estimate that cattle, sheep, and most probably all ruminant Artiodactyla, possess possibly 100 or more PAG genes, many of which are placentally expressed. The PAGs are highly diverse in sequence, with regions of hypervariability confined largely to surface-exposed loops. Selected PAG that are products of the invasive binucleate cells, expressed highly in early pregnancy at the time of trophoblast invasion and expressed weakly, if at all, in late gestation are useful in the early diagnosis of pregnancy. In a preferred embodiment, the invention relates to immunoassays for detecting these PAGs.

Les glycoprotéines associées à la grossesse (PAG) sont structurellement apparentées aux pepsines, que l'on croyait limitées aux ongulés et qui s'expriment en particulier dans la couche extérieure des cellules épithéliales (chorion/trophectoderme) du placenta. On estime que par un clonage des gènes exprimés provenant de banques génomiques placentaires d'ovins et de bovins, le bétail, les ovins, et très certainement tous les Artiodactyles ruminants, peuvent posséder au moins 100 gènes PAG, dont la plupart sont exprimés dans le placenta. Ces gènes PAG divergent largement au niveau de leurs séquences et présentent des régions d'hypervariabilité, généralement limitées à des boucles exposées en surface. Certains gènes PAG sont le produit de cellules binucléaires invasives, qui sont exprimées fortement dans une grossesse précoce au moment de l'invasion trophoblastique, et faiblement, voire pas du tout, dans une gestation tardive, ces gènes PAG permettant par ailleurs un diagnostic précoce de la grossesse. Dans un mode de réalisation préféré, cette invention concerne des essais immunologiques permettant de détecter ces gènes PAG.
