The present invention is related to the detection of bacteria, such as Mycobacteria, in human or animal body fluids such as blood, sputum and urine. The present invention provides a method for assessing the viability of bacteria such as Mycobacterium tuberculosis without the need for propagation of the bacteria. The method of the present invention is in particular useful for assessing the viability of Mycobacteria species such as are M. tuberculosis or M. leprae. With the present invention oligonucleotides are provided that can be used as primers and probes for the amplification of bacterial EF-Tu mRNA. The use of the oligonucleotides according to the invention is not limited to any particular amplification technique or any particular modification thereof. It is evident that the oligonucleotides according to the invention find their use in many different nucleic aid amplification techniques and various methods for detecting the presence of (amplified) bacterial EF-Tu mRNA.

La présente invention concerne la détection de bactéries, telles que les mycobactéries, dans des fluides organiques d'homme ou d'animal, comme le sang, les expectorations et l'urine, et elle concerne notamment un procédé d'évaluation de la viabilité de bactéries telles que les mycobactéries de la tuberculose, sans qu'il soit nécessaire de propager ces bactéries. Le procédé de l'invention est particulièrement utile pour évaluer la viabilité d'espèces de mycobactéries telles que le bacille de la tuberculose ou celui de la lèpre. Grâce à l'invention, on obtient des oligonucléotides que l'on peut utiliser en tant qu'amorces ou sondes destinées à l'amplification de l'ARNm bactérien de EF-Tu. L'utilisation d'oligonucléotides selon l'invention ne se limite pas à une quelconque technique d'amplification ou à une quelconque modification particulière de ceux-ci. Il est évident que les oligonucléotides de l'invention sont utiles dans bien des techniques différentes d'amplification d'acides nucléiques et dans plusieurs procédés de détection de la présence d'ARN bactérien (amplifié) de EF-Tu.
