A method of improving amplification of nucleic acids using a nucleic acid sequence-based amplification ("NASBA") method is provided wherein target nucleic acids and NASBA primers are electronically addressed to electronically addressable capture sites of a microchip. This improvement uses electronically induced hybridization of the target nucleic acids to the primers. The primers may be solution-based or immobilized on the capture sites of the microchip.
L'invention se rapporte à une méthode permettant d'améliorer l'amplification d'acides nucléiques et fondée sur une méthode d'amplification d'acide nucléique par séquence ("NASBA" ou nucleic acid sequenced-based amplification). Selon la méthode de cette invention, les acides nucléiques cibles et les amorces NASBA font l'objet d'un adressage électronique sur des sites de capture d'une microplaquette, susceptibles d'être adressés électroniquement. L'amélioration consiste en la mise en oeuvre d'une induction électronique de l'hybridation des acides nucléiques cibles avec les amorces. Ces amorces peuvent se présenter en solution ou être immobilisées sur les sites de capture de la microplaquette.
