The present invention relates to the electrophoretic separation of bio-organic molecules using a slab gel electrophoresis apparatus having a pair of spaced, confronting plates defining a multi-lane separation zone adapted to hold a separation medium and an upper loading zone. In one aspect of the invention, at least one of the plates is shaped to provide an expanded loading zone. The plate-to-plate distance within the expanded loading zone is greater than the plate-to-plate width within the separation zone. In another aspect of the invention, a comb is provided with each tooth having a gradual taper beginning at a support member and extending along most of the tooth's longitudinal axis, and a sharp taper beginning immediately beyond the gradual taper and extending to the end of the tooth. In a related aspect of the invention, a method is provided wherein a comb is inserted into the loading zone so that the teeth penetrate an upper portion of the separation medium, thereby forming a series of fluid-tight, elongate sample-loading apertures of substantially rectangular cross-section having a plate-to-plate width greater than that of the inter-plate distance in the separation zone.
Selon cette invention, un gel en plaque destiné à un appareil d'électrophorèse (12) comprend une paire de plaquettes opposées (14, 16), espacées l'une de l'autre et définissant une zone de séparation (22) à plusieurs voies, cette dernière étant conçue pour contenir un substrat de séparation ou un gel (23), ainsi qu'une zone supérieure de chargement. Au moins une des plaquettes est configurée pour former une zone de chargement élargie (16a-d). La distance entre les plaquettes à l'intérieur de la zone de chargement élargie est plus grande que dans la zone de séparation. Un peigne (24) possède des dents (28) dont chacune s'effile graduellement depuis un élément de support (32) sur presque toute la longueur de la dent. L'effilement de la dent est plus abrupt entre la partie à effilement graduel et le bout de la dent. On insère le peigne dans la zone de chargement de manière à ce que les dents pénètrent dans une partie supérieure du gel formant ainsi une série d'ouvertures allongées, étanches aux fluides et servant au chargement d'échantillons; ils ont une coupe sensiblement rectangulaire et sont plus larges entre les plaquettes que dans la zone de séparation. Cela permet de faire fonctionner 96 voies, même avec des gels ultraminces.
