To gain a better understanding of tumor angiogenesis, new techniques for isolating endothelial cells (ECs) and eval- uating gene expression patterns were developed. When transcripts from ECs derived from normal and malignant colorectal tissues were compared with transcripts from non-endothelial cells, over 170 genes predominantly expressed in the endothelium were iden- tified. Comparison between normal- and tumor-derived endothelium revealed 79 differentially expressed genes, including 46 that were specifically elevated in tumor-associated endothelium. Experiments with representative genes from this group demonstrated that most were similarly expressed in teh endothelium of primary lung, breast, brain, and pancreatic cancers as well as in metastatic lesions of the liver. These results demonstrate that neoplastic and normal endothelium in humans are distinct at the molecular level, and have significant implications for the development of anti-angiogenic therapies in the future.

Afin de mieux comprendre l'angiogenèse tumorale, de nouvelles techniques permettant d'isoler des cellules endothéliales et d'évaluer des profils d'expression génique ont été développées. La comparaison effectuée entre des transcrits issus de cellules endothéliales provenant de tissus colorectaux normaux et malins et des transcrits issus de cellules non endothéliales a permis d'identifier plus de 170 gènes qui sont exprimés de manière prédominante dans l'endothélium. La comparaison entre des endothéliums d'origine normale et d'origine maligne a permis de mettre en évidence 79 gènes à expression différentielle, dont 46 étaient particulièrement élevés dans des endothéliums à association tumorale. Des expériences menées avec des gènes représentatifs de ce groupe ont montré que la plupart de ces gènes étaient exprimés de manière identique dans l'endothélium de cancers primitifs du poumon, du sein, du cerveau et du pancréas, ainsi que dans des lésions métastatiques du foie. Ces résultats montrent que l'endothélium néoplasique et l'endothélium normal sont distincts chez l'homme, au niveau moléculaire, et sont sensiblement impliqués dans le développement de futures thérapies anti-angiogéniques.
