Mutant glycosidases in which the amino acid in the active site that serves as the acid, base or acid/base-catalyst is converted from a carboxylic acid to some other amino acid (for example to a simple alkyl, as in alanine or glycine) can catalyze the reaction of a thiosugar acceptor and an activated donor to form a thioglycoside. The "thioglycoligases" represent a novel class of mutant enzymes, and represent a first aspect of the invention. Thioglycoligases can be used in accordance with the method of the invention to couple a thiosugar acceptor and an activated donor to form a thioglycoside. By selection of the donor and acceptor species, as well as the specific enzyme employed, thioglycosides of different structure and stereochemistry can be obtained.
Des glycosidases mutantes dans lesquelles l'acide aminé dans le site actif servant de catalyseur acide, base ou acide/base est converti à partir d'un acide carboxylique en un autre acide aminé (par exemple un alkyle simple, comme dans l'alanine ou la glycine) peuvent catalyser la réaction d'un accepteur de thio-sucres et d'un donneur activé pour former un thioglycoside. Les "thioglycoligases" constituent une nouvelle classe d'enzymes mutantes et font l'objet d'un premier aspect de l'invention. Les thioglycoligases peuvent être utilisées d'après le procédé selon l'invention pour coupler un accepteur de thio-sucres et un donneur activé de façon à former un thioglycoside. Par sélection de l'espèce du donneur et de l'espèce de l'accepteur et de l'enzyme spécifique employée, des thioglycosides présentant des structures et des stéréochimies différentes peuvent être obtenus.
