Stable clones of neural stem cells (NSCs) have been isolated from human fetal telencephalon. In vitro, these self-renewing clones can spontaneously give rise to all three fundamental neural cell types (neurons, oligodendrocytes and astrocytes). Following transplantation into germinal zones of the developing newborn mouse brain, these NSCs, like their rodent counterparts can participate in aspects of normal development. These include migration along well-established migratory pathways to disseminated CNS regions, differentiation into developmentally and regionally appropriate cell types in response to microenvironmental cues and non-disruptive, non-tumorigenic interspersion with host progenitors.
L'invention concerne des clones stables de cellules neuronales souches isolées du télencéphale foetal humain. In vitro, ces clones auto-renouvelés peuvent produire spontanément tous les trois types fondamentaux de cellules neuronales (à savoir neurones, oligodendrocytes et astrocytes). Suite à une transplantation dans des zones germinatives du cerveau en développement d'un souriceau nouveau-né, ces cellules neuronales souches peuvent participer, à l'instar de leurs homologues chez les rongeurs, à certains aspects de développement normal tels que la migration le long des voies de migration bien établies vers des régions disséminées du SNC, la différentiation en des types de cellules appropriés des points de vue du développement et de zones en réponse à des indices du micro-environnement et, enfin, la dispersion sans perturbations ni effets tumorigènes parmi les progéniteurs hôtes.
