Primer compositions for anti-cancer agent delivery to solid tumors in mammals, including man, are disclosed. The primers comprise a pharmaceutically acceptable agent which increases the intracellular levels of cyclic adenosine monophosphate (cAMP) in the normal connective tissue cells within solid tumors. Such an agent may be selected from any agents which increase the intracellular levels of cAMP, e.g. agents binding to and stimulating receptors at the connective tissue cell surface within tumors, and agents inhibiting cAMP phosphodiesterase. Local administration of the primer to the solid tumor area results in a lowering of the interstitial pressure. Further, an agent delivery system and a method for delivery of anti-cancer agents to solid tumors are described.
L'invention concerne des compositions d'amorces prévues pour être administrées dans des tumeurs solides, lors d'un traitement par agents anticancéreux, chez le mammifère, y compris chez l'homme. Ces amorces comprennent un agent pharmaceutiquement acceptable qui permet d'augmenter les concentrations intracellulaires d'adénosine-monophosphate cyclique dans les cellules normales des tissus conjonctifs, à l'intérieur des tumeurs solides. Un agent de ce type peut être sélectionné parmi des agents qui élèvent les concentrations intracellulaires d'adénosine-monophosphate cyclique, par exemple, des agents de liaison et de stimulation des récepteurs au niveau de la surface cellulaire des tissus conjonctifs dans les tumeurs, et des agents inhibant la phosphodiestérase de l'adénosine-monophosphate cyclique.
