Adding at least one gene involved in plant host cell T-DNA integration enhances transformation by Agrobacterium. The histone H2A gene encoded by the Arabidopsis RAT5 gene increases transformation frequencies of plants, most likely by causing overexpession of a product needed for T-DNA integration. Agrobacterium tumefaciens genetically transforms plant cells by transferring a portion of the bacterial Ti-plasmid, designated the T-DNA, to the plant, and integrating the T-DNA into the plant genome. However, not all plants are transformable by Agrobacterium and transformation frequencies may be too low to be useful. Little is known about the T-DNA integration process, and no plant genes involved in integration have been identified prior to the present invention.

L'invention concerne l'adjonction d'au moins un gène intervenant dans l'intégration de l'ADN-T de cellules hôtes de plante, qui améliore la transformation par <i>Agrobacterium</i>. Le gène H2A histone codé par le gène RAT5 <i>Arabidopsis</i> augmente les fréquences de transformation des plantes, le plus vraisemblablement par surexpression d'un produit requis pour l'intégration de l'ADN-T. <i>Agrobacterium tumefaciens</i> transforme génétiquement les cellules de plante par transfert d'une partie de plasmide Ti bactérien, appelé ADN-T, à la plante, et par intégration de cet ADN-T dans le génome de la plante. Néanmoins, toutes les plantes ne sont pas transformables par <i>Agrobacterium</i>, et les fréquences de transformation peuvent être trop faibles pour être utiles. On sait peu de choses sur le processus d'intégration de l'ADN-T, et aucun gène de plante intervenant dans cette intégration n'a été identifié avant la présentation de l'invention.
