The present invention is directed to methods and compositions for using DNA analog probes in increasing the efficiency of DNA targeting by recombinase coated nucleoprotein filaments. The present invention teaches novel methods and compositions that combine the traditional advantages of RecA coated filaments in catalyzing homology searches with the utility of PNA (peptide nucleic acids) to bind DNA with a very high affinity and its ability to locally open the target DNA thereby improving the kinetics of RecA-mediated strand exchange.
L'invention concerne des procédés et des compositions servant à mettre en application des sondes d'analogues d'ADN afin d'augmenter l'efficacité du ciblage de l'ADN par des filaments de nucléoprotéines revêtus par recombinase. Elle concerne de nouveaux procédés et de nouvelles compositions combinant les avantages classiques de filaments revêtus de RecA dans la catalyse de recherches d'homologie et la capacité des ANP (acides nucléiques peptidiques) de fixation à l'ADN avec une affinité très élevée et d'ouverture localisée de l'ADN ciblé, ce qui permet d'améliorer le potentiel cinétique de l'échange de brins dans lequel RcA joue un rôle.
