T-cell responses are initiated via contact with MHC/peptide complexes on antigen presenting cells (APCs). The fate of these complexes, however, is unknown. Here, using live APCs expressing MHC class I molecules fused with green-fluorescent protein, we show that peptide-specific T-cell/APC interaction induces clusters of MHC I molecules to congregate within minutes at the contact site; thereafter, these MHC I clusters are acquired by T-cells in small aggregates. We further demonstrate that acquisition of MHC I by T-cells correlates with TCR down regulation and the APC-derived MHC I molecules are endocytosed and degrated by T-cells. These data suggest a novel mechanism by which TCR recognition of MHC/peptide complexes can be curtailed by internalization of MHC molecules by T-cells.

Les réponses des lymphocytes T surgissent de leur mise en contact avec des complexes MHC/peptide sur les cellules présentatrices de l'antigène (CPA). Le sort de ces complexes est toutefois inconnu. L'invention a pour objectif de démontrer, par le biais des molécules MHC de classe I exprimant des CPA vivantes fusionnées avec une protéine fluorescente verte, que l'interaction entre les lymphocytes T et les CPA spécifiques du peptide induit la formation de groupes de molécules MHC I se produisant en quelques minutes au site de contact. Les groupes MHC I sont, ensuite, acquises par les lymphocytes T dans de petits groupes. Elle vise aussi à démontrer que l'acquisition de MHC par des lymphocytes T est en relation avec la régulation à la baisse de TCR et que les molécules MHC I issues des CPA sont endocytosées et dégradées par les lymphocytes T. Ces données laissent supposer un nouveau mécanisme grâce auquel on peut résorber la reconnaissance TCR des complexes MHC/peptide en internationalisant les molécules MHC par les lymphocytes T.
