This invention is directed to an optically-based method and system for analyte detection using solid phase immobilization, specific analyte labels adapted for signal generation and corresponding processes for the utilization thereof. The enumeration detection method disclosed herein narrows the area for signal observation, thus, improving detectable signal to background ratio. The system is comprised of a platform/support for immobilizing a sample stage having a labeled sample (analyte complex) bound thereto, a radiation source, an optical apparatus for generating and directing radiation at said sample and a means for data collection and analysis. Upon engagement of the system, the sample generates a signal capable of differentiation from background signal, both of which are collected and imaged with a signal detector that generated a sample image to a data processing apparatus. Said apparatus receives signal measurements and, in turn, enumerates individual binding events. Generated signal may be increased via selected mass enhancement. The invention, enumeration assay methodology detecting individual binding events, may be used, for example, in analyses to detect analyte or confirm results in both research, commercial and point of care applications.
La présente invention concerne un procédé et un système optiques permettant de détecter un analyte par une immobilisation en phase solide, des marqueurs d'analyte spécifiques aptes à produire un signal et des procédés d'utilisation de ces derniers. Le procédé de détection par numération de l'invention permet de rétrécir la zone d'observation du signal et, par conséquent, d'améliorer le rapport signal détectable/fond. Le système comprend une plate-forme/support permettant d'immobiliser une platine d'échantillon sur laquelle est lié un échantillon marqué (complexe d'analyte), une source de rayonnement, un appareil optique destiné à générer et à diriger le rayonnement vers l'échantillon et un moyen de collecte et d'analyse des données. Au moment où le système est enclenché, l'échantillon génère un signal pouvant se différencier du signal de fond, les deux signaux étant collectés et mis en image grâce à un détecteur de signal qui transmet une image de l'échantillon à un appareil de traitement des données. Ledit appareil reçoit les mesures du signal et, à son tour, effectue la numération des événements de liaison individuels. Il est possible de renforcer le signal généré par une augmentation choisie de la masse. Le procédé d'analyse par numération de l'invention, qui permet de détecter des événements de liaison individuels, peut être utilisé, par exemple, dans des analyses visant à détecter un analyte ou à confirmer des résultats dans des applications de recherche, des applications commerciales ou aux endroits où des soins sont dispensés.
