The invention provides a method of producing a polypeptide having a C-terminal .alpha.-carboxamide group. It particularly concerns an enzymatic modification of selected substrate polypeptides which result in cleavage of the substrate polypeptide to form a product peptide with a C-terminal arginine residue having an .alpha.-carboxamide group (C-terminal "Arg-NH2"). The method includes contacting an aqueous-based solution including (i) ammonia reagent and (ii) the substrate polypeptide with (iii) clostripain.
L'invention concerne un procédé permettant de produire un polypeptide présentant un groupe .alpha.-carboxamide C-terminal. Elle concerne en particulier une modification enzymatique de polypeptides substrats choisis qui produit un clivage du polypeptide substrat pour former un produit peptidique avec un résidu C-terminal arginine présentant un groupe .alpha.-carboxamide (C-terminal "Arg-NH¿2?"). Le procédé consiste en outre à mettre en contact une solution à base aqueuse comprenant (i) un réactif d'ammoniac et (ii) le polypeptide substrat avec (iii) du clostripain.
