ABSTRACT ENZYME ASSAY METHOD A method for the assay of an enzyme comprises incubatiing a sample containing the enzyme with a reagent comprising a substrate for the enzyme which reacts with the enzyme to give rise to the formation of a compound which is monitored, characterised in that said compound is of formula HX-A-Y-NO2, wherein A represents an aromatic nucleus, HX represents an auxochromic group, preferably a hydroxyl group, and Y represents an unsaturated group which is capable of transmitting electron resonance between the aromatic nucleus (A) and the nitro substituent (NO2), said compound being capable of producing a visual signal which is discernible by eye either directly or on alkaline treatment. Preferably the reagent comprises an enzyme substrate of formula S'-X-A-Y-NO2, X represents an auxochromic radical corresponding to an auxochromic group HX and S' represents an appropriate carboxylate, phosphate, diphosphate, sulphate or glycoside residue, according to the nature of the enzyme being assayed, e.g. carboxylate esterase or lipase, acid or alkaline phosphotase, phophodiesterase, sulphatase or glycosidase.
