This invention provides novel substrate compounds that selectively inhibit the proliferation of pathological cells, for example, pathological cells that endogenously overexpress a target enzyme that confers resistance to biologic and chemotherapeutic agents. The enzyme acts on a substrate compound to 1) convert it to a cellular toxin and/or 2) release a toxic byproduct. In one embodiment, the activity of the target enzyme has been greatly enhanced in a target cell as a result of loss of tumor suppressor function and/or selection resulting from previous exposure to chemotherapy. In another embodiment, the pathological cell contains a target enzyme that is an expression product of an infectious agent in the cell. Further provided by this invention is a method for treating a subject by delivering to the subject a prodrug as described herein. The prodrugs of this invention may be used alone or in combination with other chemotherapeutics or alternative anti-cancer therapies such as radiation.
L'invention concerne de nouveaux composés substrats qui inhibent de manière sélective la prolifération de cellules pathologiques, par exemple, de cellules pathologiques qui surexpriment de manière endogène un enzyme cible qui confère une bonne résistance aux agents biologiques et chimiothérapeutiques. L'enzyme agit sur un composé substrat de manière 1) à le convertir en une toxine cellulaire et/ou 2) à libérer un sous-produit toxique. Dans un mode de réalisation, l'activité de l'enzyme cible a été considérablement améliorée dans une cellule cible en raison de la perte de la fonction de suppression des tumeurs et/ou de la sélection résultant d'une exposition préalable à un traitement chimiothérapeutique. Dans un autre mode de réalisation, la cellule pathologique contient un enzyme cible qui est un produit d'expression d'un agent infectieux dans la cellule. L'invention concerne également une méthode de traitement qui consiste à administrer à un patient un promédicament tel que décrit par l'invention. Les promédicaments selon l'invention peuvent être utilisés seuls ou combinés à d'autres thérapies chimiothérapeutiques ou à d'autres thérapies contre le cancer telles que la radiothérapie.
