This invention provides a method for identifying potential therapeutic agents by contacting a target cell with a candidate therapeutic agent which is a selective substrate for an endogenous, intracellular enzyme in the cell which is enhanced in its expression as a result of selection by biologic or chemotherapy. This invention also provides methods and examples of molecules for selectively killing a pathological cell by contacting the cell with a prodrug that is a selective substrate for an endogenous, intracellular enzyme. The prodrug is subsequently converted to a cellular toxin. Further provided by this invention is a method for treating a pathology characterized by pathological, hyperproliferative cells in a subject by administering to the subject a prodrug that is a selective substrate for an endogenous, overexpressed, intracellular enzyme, and converted by the enzyme to a cellular toxin in the hyperproliferative cell.
L'invention se rapporte à une méthode d'identification d'agents thérapeutiques potentiels consistant à mettre en contact une cellule cible avec un agent thérapeutique candidat qui est un substrat sélectif pour un enzyme intracellulaire et endogène qui est présent dans la cellule et dont l'expression est accentuée du fait d'une sélection par processus biologique ou chimiothérapeutique. L'invention se rapporte également à des procédés et à des exemples de molécules permettant de détruire sélectivement une cellule pathologique par mise au contact de ladite cellule avec un bioprécurseur qui est un substrat sélectif pour un enzyme endogène, intracellulaire. Ledit bioprécurseur est ensuite converti en une toxine cellulaire. L'invention se rapporte également à une méthode permettant de traiter une pathologie caractérisée par des cellules pathologiques, hyperprolifératives, chez un sujet et consistant à administrer audit sujet un bioprécurseur qui est un substrat sélectif pour un enzyme intracellulaire, surexprimé, endogène et converti par l'enzyme en une toxine cellulaire dans la cellule hyperproliférative.
