The present invention relates to methods and compositions designed for the treatment, management, or prevention of cancer, particularly, metastatic cancer. In one embodiment, the methods of the invention comprise the administration of an effective amount of an antibody that binds to EphA2 and agonizes EphA2, thereby increasing EphA2 pho 3phorylation and decreasing EphA2 levels. In other embodiments, the methods of the invention comprise the administration of an effective amount of an antibody that binds to Eph A2 and inhibits cancer cell colony formation in soft agar, inhibits tubular network fore cation in three-dimensional basement membrane or extracellular matrix preparation, preferentially binds to an EphA2 epitope that is exposed on cancer cells but not non-cancer cells, and/or has a low Koff, thereby, inhibiting tumor cell growth and/or metastasis. The invention also provides pharmaceutical compositions comprising one or more EphA2 antil)odies of the invention either alone or in combination with one or more other agents useful for cancer therapy.

L'invention concerne des méthodes et des compositions conçues pour traiter, prendre en charge ou prévenir le cancer, notamment le cancer métastatique. Dans un mode de réalisation, les méthodes de l'invention consistent à administrer une quantité efficace d'un anticorps qui se lie à EphA2 et agonise EphA2, ce qui permet d'augmenter la phophorylation de EphA2 et de diminuer les niveaux de EphA2. Dans d'autres modes de réalisation, les méthodes de l'invention consistent à administrer une quantité efficace d'un anticorps qui se lie à EphA2, inhibe la formation de colonies de cellules cancéreuses dans la gélose molle, inhibe la formation d'un réseau tubulaire dans une membrane basale tridimensionnelle ou la préparation d'une matrice extracellulaire, se lie de préférence à un épitope de EphA2 exposé sur des cellules cancéreuses et non sur des cellules non cancéreuses et/ou présente un faible K¿o?ff, ce qui permet d'inhiber la croissance de cellules tumorales et/ou des métastases. L'invention concerne également des compositions pharmaceutiques comprenant un ou plusieurs anticorps contre EphA2 soit seul(s) soit en combinaison avec un ou plusieurs autres agents utilisés dans une thérapie cancéreuse.
