Antibodies to two new epitopes on the HCV envelope proteins were identified which allow routine detection of native HCV envelope antigens, in tissue or cells derived from the host. The new epitopes are: the E1 region aa 307-326 and the N-terminal hyper variable region of E2 aa 395-415. Surprisingly, we characterised an antibody that reacts with various sequences of the hypervariable domain of E2. Specific monoclonal antibodies directed against these epitopes and allowing routine detection of viral antigen are described.
L'invention porte sur deux nouveaux épitopes des protéines enveloppes du VHC (virus de l'hépatite C) permettant la détection courante d'antigènes natifs enveloppe du VHC dans des tissus ou des cellules provenant de l'hôte. Ces nouveaux épitopes sont la région aa 307-326 de la E1 et la région N-terminale hypervariable aa 395-415 de la E2. On a eu la surprise de découvrir un anticorps réagissant avec différentes séquences du domaine hypervariable de la E2. L'invention porte également sur des anticorps monoclonaux spécifiques de ces épitopes, et la détection courante d'antigènes viraux.
