EUKARYOTIC AUTONOMOUSLY REPLICATING SEGMENT ABSTRACT OF THE DISCLOSURE Method, compositions and microorganisms involving host eukaryotic chromosomal DNA providing autonomous repli- cating segments (ars) joined to at least one gene capable of expression and replication in a eukaryotic host not naturally joined to the ars. A eukaryotic chromosome is segmented and the segments joined to a gene capable of expression in a yeast and having a phenotypic marker allowing for selection to provide a hybrid DNA molecule. Yeast are transformed with the hybrid DNA molecule and cultured under conditions select- ing for the transformants, whereby the hybrid DNA molecules may be isolated providing a source of the autonomous repli- cating segment. The autonomous replicating segment may then be used in conjunction with one or more genes to transform the host or other eukaryotes to provide for multiple copies of a host gene, an exogenous gene, or for integration or recombination with the host chromosome, as well as expression of the gene(s). This invention was developed under grants from the National Science Foudation, National Instiute of Health and the United States Public Health Service.
