The invention provides a cell composition comprising a population of non-yeast eukaryotic cells containing a diverse population of variant nucleic acids, each of the variant nucleic acids being expressed in a different cell and located within each cell at an identical site in the genome. The invention also provides a method of identifying a polypeptide exhibiting optimized activity by sceening a population of non-yeast eukaryotic cells containing a diverse population of variant nucleic acids for an activity associated with a parent polypeptide of a diverse population of variant polypeptides encoded by the variant nucleic acids; and identifying a variant polypeptide exhibiting an optimized activity relative to the parent polypeptide.

L'invention concerne une composition cellulaire qui comprend une population de cellules eucaryotes sans levure contenant une population différenciée d'acides nucléiques variants, chaque acide nucléique variant étant exprimé dans une cellule différente et située à l'intérieur de chaque cellule à un emplacement identique dans le génome. L'invention concerne également une méthode d'identification d'un polypeptide présentant une activité optimisée, qui consiste à cribler une population de cellules eucaryotes sans levure contenant une population différenciée d'acides nucléiques variants pour une activité associée à un polypeptide parent d'une population différenciée de polypeptides variants codés par les acides nucléiques variants. La méthode consiste ensuite à identifier un polypeptide variant présentant une activité optimisée relativement au polypeptide parent.
