This invention provides compositions and methods for producing translational components that expand the number of genetically encoded amino acids in eukaryotic cells. The components include orthogonal tRNAs, orthogonal aminoacyl-tRNA synthetases, orthogonal pairs of tRNAs/synthetases and unnatural amino acids. Proteins and methods of producing proteins with unnatural amino acids in eukaryotic cells are also provided.

Cette invention concerne des compositions et des méthodes de production de composants traductionnels qui augmentent le nombre d'acides aminés codés génétiquement dans des cellules eucaryotes. Ces composants comprennent des ARNt orthogonaux, des aminoacyl-ARNt synthétases orthogonales, des paires orthogonales d'ARNt synthétases et des acides aminés artificiels. L'invention concerne en outre des protéines et des méthodes de production de protéines présentant des acides aminés artificiels dans des cellules eucaryotes.
