This invention relates, e.g., to a method for expanding mammalian acinar cells, comprising culturing the cells in a cell culture system comprising a cell culture medium and a cell attachment surface, under conditions wherein the acinar cells undergo a 3-4 fold expansion together with transdifferentiation into a modified cell phenotype (IP cells) showing characteristics of acinar cells and liver cells. The invention also relates to a method for transforming these IP cells to insulin-producing cells in vitro, comprising culturing the cells in a novel, defined medium. Also disclosed are suitable culture media for performing these methods, isolated cells having the phenotype of IP cells and/or produced by these methods, and kits for performing the methods.

L'invention concerne, par exemple, un procédé destiné à l'expansion des cellules acineuses mammaliennes qui consiste à cultiver les cellules dans un système de culture de cellules comprenant un milieu de culture de cellule et une surface de fixation de cellules, dans des conditions où les cellules acineuses grossissent de 3 à 4 fois et subissent une transdifférentiation dans un phénotype de cellules modifié (cellules IP) manifestant des caractéristiques des cellules acineuses et des cellules du foie. L'invention concerne aussi un procédé pour transformer ces cellules IP en cellules productrices d'insuline in vitro, qui consiste à cultiver les cellules dans un nouveau milieu défini. Elle concerne également des milieux de culture pour mettre en oeuvre ce procédé, des cellules isolées possédant le phénotype des cellules IP et/ou produites par ces procédés ainsi que des kits pour mettre en oeuvre ces procédés.
