HBsAg is expressed in a Saccharomyces cerevisiae host, carrying a plasmid having a HBsAg coding sequence, wherein the plasmid includes: (1) an upstream promoter from a glyceraldehyde-3- phosphate dehydrogenase gene, for controlling expression of the HBsAg coding sequence; and (2) an ARG3 transcription terminator downstream of the HBsAg coding sequence. The plasmids may also include: (3) a LEU2 selection marker; (4) a 2µ plasmid sequence; and (5) an origin of replication functional in Escherichia coli. HBsAg can be expressed in this host, and can be purified for use in the manufacture of vaccines, and in particular for the manufacture of combination vaccines and in new monovalent HBV vaccines e.g. with non-alum adjuvants.
Selon la présente invention, HbsAg est exprimé dans un hôte Saccharomyces cerevisiae portant un plasmide qui présente une séquence de codage de HbsAg. Ce plasmide comprend (1) un promoteur amont provenant d'un gène de glycéraldéhyde-3-phosphate déshydrogénase, conçu pour commander l'expression de la séquence de codage de HbsAg, et (2) un terminateur de transcription d'ARG3, en aval de la séquence de codage de HbsAg. Les plasmides peuvent également comprendre (3) un marqueur de sélection LEU2, (4) une séquence de plasmide 2µ et (5) une origine de réplication fonctionnelle dans Escherichia coli. HbsAg peut être exprimé dans cet hôte et peut être purifié pour être utilisé afin de produire des vaccins, en particulier pour produire des vaccins combinés et dans de nouveaux vaccins anti HBV monovalents, par exemple avec des adjuvants sans alun.
