The invention relates to a method for analysing a genomicregion of an organism, comprising four major parts. The first part involves the isolation of mRNA from a selected organism that is used for the preparation of small single stranded DNA fragments with one adaptor containing an affinity label. These DNA fragments are used in part three. In the second part genomic DNA from the same or a related organism is isolated. This genomic DNA is fragmented and ligated to adaptor molecules. In the third part these genomicfragments are hybridized with single stranded DNA fragments from part one and the hybrids formed inthis process are used for synthesis of DNA fragments. These fragments will be used in part four, which involves sequencing of these fragments using one of the available high throughput sequencing methods.
L'invention concerne un procédé d'analyse d'une microrégion génomique d'un organisme, comportant quatre parties principales. La première partie implique l'isolement dARNm à partir d'un organisme sélectionné utilisé pour la préparation de petits fragments d'ADN à simple brin avec un adaptateur contenant un marqueur d'affinité. Ces fragments d'ADN sont utilisés dans la troisième partie. Dans la deuxième partie, on isole l'ADN génomique du même organisme ou d'un organisme apparenté. On fragmente cet ADN génomique et on le ligature à des molécules adaptatrices. Dans la troisième partie, on hybride ces fragments génomiques avec des fragments d'ADN à simple brin de la première partie et on utilise les hybrides formés dans ce processus pour la synthèse des fragments d'ADN. Ces fragments seront utilisés dans la quatrième partie qui implique le séquençage de ces fragments à l'aide de l'une des méthodes de séquençage à haut débit disponibles.
