The present invention relates to the general field of recombinant protein expression, purification of recombinant proteins, diagnosis of HCV infection, prophylactic treatment against HCV infection and to the prognosing/monitoring of the clinical efficiency of treatment of an individual with chronic hepatitis, or the prognosing/ monitoring of the natural disease. In particular, the present invention relates to the use of yeast, i.e. Hansenula or Saccharomyces glycosylation minus strains, for the efficient expression of HCV envelope proteins that are core-glycosylated, purification methods for these proteins, and the use in various applications, such as the use in diagnosis, prophylaxis or therapy of HCV envelope proteins purified according to the present invention.
L'invention concerne le domaine général de l'expression des protéines de recombinaison, la purification de ces protéines, le diagnostic de l'infection par le VHC, le traitement prophylactique contre ladite infection, et le pronostic/l'évaluation de l'efficacité clinique du traitement appliqué à un patient souffrant d'hépatite chronique, ou bien le pronostic/l'évaluation de la maladie naturelle. En particulier, l'invention concerne l'utilisation de la levure, c'est-à-dire de souches négatives de glycosylation Hansenula ou Saccharomyces, pour l'expression efficace de protéines de l'enveloppe du VHC à nucléo-glycosylation, mais elle concerne également des procédés de purification pour ces protéines, et l'utilisation, pour différentes applications (par exemple, diagnostic, prophylaxie ou thérapie), de protéines de l'enveloppe du VHC purifiées selon les modalités décrites dans l'invention.
