A method for producing a polypeptide product which is substantially free of an undesired protein, the process comprising culturing a host cell which is able to express said polypeptide product and which is able to express said undesired protein only in a mutant form which form has the activity of the corresponding native protein under culture conditions but is unstable under conditions at which the said polypeptide product remains stable; and recoverig the desired product, wherein either the host cell culture or the recovered product is subjected for a sufficient period of time to conditions under which the undesired protein is unstable so as to denature the undesired protein. The method allows the production of products which are substantially free of particular undesired contaminants.

L'invention concerne un procédé d'obtention d'un produit polypeptidique pratiquement exempt d'une protéine indésirable. Ce procédé consiste à cultiver une cellule hôte capable d'exprimer d'une part ledit produit polypeptidique, d'autre part la protéine indésirable uniquement sous forme mutante, forme dont l'activité correspond à la protéine native dans des conditions de culture, mais qui est instable dans les conditions où ledit produit polypeptidique reste stable. On récupère ensuite ledit produit, la culture de la cellule hôte ou le produit récupéré étant soumis pendant un laps de temps suffisant aux conditions dans lesquelles la protéine indésirable est instable au point de se dénaturer. Ce procédé permet d'obtenir des produits qui sont plus ou moins exempts de certains contaminants.
