Exocyclic peptide mimetics that disable Fas were developed. A three dimensional model of the Fas receptor-ligand complex was constructed and structurally predicted regions of the receptor that were relevant to binding ligand were used to create constrained peptide mimetics. Exocyclic anti-Fas peptide mimetics were identified that block Fas receptor-ligand interactions, and modulate Fas biological activity both in vitro and in vivo. The mimetics are useful, e.g., for treating Fas-related pathologies.
L'invention concerne des mimétiques peptidiques exocycliques qui désactivent Fas. On a construit un modèle à trois dimensions du complexe ligand-récepteur de Fas et on a utilisé des régions du récepteur structurellement prédites, pertinentes au ligand de liaison, pour créer des mimétiques peptidiques contraints. On a identifié des mimétiques peptidiques anti-Fas exocycliques qui bloquent les interactions ligand-récepteur de Fas et modulent l'activité biologique de Fas aussi bien in vitro qu'in vivo. Les mimétiques servent, par exemple, dans le traitement de pathologies liées à Fas.
