The present invention relates to antibodies or fragments thereof that specifically bind Fc.gamma.RIIB, particularly human Fc.gamma.RIIB, with greater affinity than said antibodies or fragments thereof bind Fc.gamma.RIIA, particularly human Fc.gamma.RIIA. The present invention also provides the use of an anti-Fc.gamma.RIIB antibody or an antigen-binding fragment thereof, as a single agent therapy for the treatment, prevention, management, or amelioration of a cancer, preferably a B-cell malignancy, particularly, B-cell chronic lymphocytic leukemia or non-Hodgkin's lymphoma, an autoimmune disorder, an inflammatory disorder, an IgE-mediated allergic disorder, or one or more symptoms thereof. The invention provides methods of enhancing the therapeutic effect of therapeutic antibodies by administering the antibodies of the invention to enhance the effector function of the therapeutic antibodies. The invention also provides methods of enhancing efficacy of a vaccine composition by administering the antibodies of the invention.
La présente invention concerne des anticorps ou leurs fragments qui se lient spécifiquement au Fc.gamma.RIIB, en particulier au Fc.gamma.RIIB humain, avec une affinité plus grande que celle avec laquelle lesdits anticorps ou leurs fragments se lient au Fc.gamma.RIIA, en particulier au Fc.gamma.RIIA humain. L'invention concerne également l'utilisation d'un anticorps anti-Fc.gamma.RIIB, ou d'un de ses fragments se liant à l'antigène, en tant que monothérapie pour le traitement, la prévention, la gestion ou l'amélioration d'un cancer, de préférence une tumeur maligne à cellules B, en particulier une leucémie lymphoïde chronique à cellules B ou un lymphome non hodgkinien, une affection auto-immune, un trouble inflammatoire, une affection allergique à réponse IgE ou un ou plusieurs de leurs symptômes. L'invention concerne des procédés permettant d'améliorer l'effet thérapeutique des anticorps thérapeutiques en administrant les anticorps de l'invention pour améliorer la fonction effectrice des anticorps thérapeutiques. L'invention concerne également des procédés d'amélioration de l'efficacité d'une composition vaccinale par l'administration des anticorps de l'invention.
