Previous methods for culturing human embryonic stem cells have required either fibroblast feeder cells or a medium which has been exposed to fibroblast feeder cells in order to maintain the stem cells in an undifferentiated state. It has now been found that if an antagonist of bone morphogenic protein is added to the medium in which the stem cells are cultured, together with fibroblast growth factor, the stem cells will remain undifferentiated indefinitely, even without feeder cells or conditioned medium.
Les méthodes antérieures de culture de cellules souches embryonnaires humaines requéraient l'utilisation de cellules nourricières fibroblastiques ou d'un milieu exposé à des cellules nourricières fibroblastiques pour maintenir les cellules souches dans un état indifférencié. On a désormais découvert qu'avec l'ajout d'un antagoniste de la protéine morphogénique osseuse dans le milieu dans lequel les cellules souches sont cultivées, conjointement avec un facteur de croissance fibroblastique, les cellules souches demeurent indifférenciées indéfiniment, même sans cellules nourricières ou milieu conditionné.
