Disclosed is a finderon having the ability to endonucleolytically cleave a sequence of 3' to 5' linked ribonucleotides. The finderon includes a rigid linker comprising at least one non-nucleotidic unit, flanked by first and second flanking regions of at least four contiguous, covalently-linked nucleotides. At least a portion of each flanking region is complementary to a target region on a substrate RNA molecule. Also disclosed are methods of preparing and using a finderon, and kits including a finderon.
L'invention porte sur une nouvelle molécule appelée "finderon", apte à effectuer un clivage endonucléolytique d'une séquence de ribonucléotides à liaisons de 3' à 5'. Ce "finderon" comporte un liant rigide comprenant au minimum une unité non nucléotide, flanquée par une première et une deuxième région constituées d'au moins quatre nucléotides contigus à liaisons covalentes. Une partie au moins de chacune de ces régions est complémentaire d'une région cible sur une molécule ARN de substrat. L'invention porte également sur des méthodes d'élaboration et d'utilisation de ce "finderon" et sur des kits renfermant de tels "finderons".
