A flaviviral expression vector, construct and system are provided that each comprise a flaviviral replicon that facilitates intracellular self-replication leading to high-level expression of RNA and protein(s) encoded by a heterologous nucleic acid. The flaviviral expression vector, construct and system preferably use a Kunjin virus replicon and are of particular use as a vaccine delivery system that facilitates expression of immunogenic proteins and peptides that induce protective T cell immunity to viral infections and cancer. Vaccines may be administered in the form of DNA, RNA or virus like particles.
L'invention concerne un vecteur d'expression, un produit de synthèse et un système à flavivirus comprenant chacun un réplicon de flavivirus facilitant l'autoreproduction intracellulaire générant un haut niveau d'expression d'ARN et de protéines codées par un acide nucléique hétérologue. Le vecteur d'expression, le produit de synthèse et le système à flavivirus comprennent, de préférence, un réplicon du virus Kunjin et servent, en particulier, de système d'administration de vaccin facilitant l'expression des protéines et des peptides immunogéniques induisant l'immunité protectrice des lymphocytes T contre les infections virales et le cancer. Les vaccins peuvent être administrés sous forme de particules d'ADN, ARN ou virales.
