ABSTRACT A reagent is described which when mixed with whole blood, lyses the red cells substantially instantly, but does not substantially affect the optical, light scattering properties of the white blood cells. This reagent enables the performance of a five part leukocyte differential by flow cytometry and light scattering in a single sample path white cell analyzer. In its preferred embodiment, the lytic agent combines the functions of leukoprotective and antimicrobial agents, selected nonionic surfactants, a lytic and wetting agent and an appropriate buffer to provide pH buffering and to increase the solution's conductivity and red blood cell lysing capability. -25-
