ABSTRACT Flow-through nucleic acid hybridization assays in which ligand- derivatized probes are synthesized using a target nucleic acid sequence as the template. The probe is immobilized on the membrane of a flow-through assay device by binding to a specific binding pair member for the ligand or hybridization to a complementary nucleic acid sequence. The probe on the membrane of a flow-through assay device, indicating the template target sequence was present during probe synthesis, is detected by means of a tracer comprising a specific binding molecule for the ligand.
