A method for the fluorescent detection of a substance, the method comprising providing particles comprising a metal or a metal oxide core, wherein one or more optionally fluorescently tagged antibodies or human specific peptide nucleic acid (PNA) oligomers for binding to a substance is/are bound, directly or indirectly, to the surface of the metal or metal oxide; contacting a substrate, which may or may not have the substance on its surface, with the particles for a time sufficient to allow the antibody/PNA oligomer to bind with the substance; removing those particles which have not bound to the substrate; if the antibodies or PNA oligomers are not fluorescently tagged, contacting the substrate with one or more fluorophores that selectively bind with the antibody and/or substance, then optionally washing the substrate to remove unbound fluorophores; and illuminating the substrate with appropriate radiation to show the fluorophores on the substrate.
L'invention concerne un procédé de détection par fluorescence d'une substance. Ce procédé consiste à prévoir des particules contenant un métal ou un noyau d'oxyde métallique, au moins un anticorps éventuellement marqué par fluorescence ou oligomère d'acide nucléique peptidique (PNA) spécifique humain pour la liaison à une substance étant lié directement ou indirectement à la surface du métal ou de l'oxyde métallique; à mettre en contact un substrat, présentant ou non la substance sur sa surface, avec les particules pendant un temps suffisamment long pour permettre à l'anticorps/oligomère PNA de se lier à la substance; à retirer les particules qui ne se sont pas liées au substrat; si les anticorps ou les oligomères PNA ne sont pas marqués par fluorescence, à mettre en contact le substrat avec au moins un fluorophore qui se lie sélectivement à l'anticorps et/ou à la substance, puis éventuellement à laver le substrat pour retirer les fluorophores non liés; et enfin à éclairer le substrat en l'exposant à des rayonnements appropriés pour mettre en évidence les fluorophores sur le substrat.
