Disclosed is a method for increasing the fluorescence of a Cyanine dye molecule comprising at least one NO2 group characterised by the reduction of the at least one NO2 group to NHOH or NH2 by the action of a nitroreductase. The cyanine dye molecule comprising at least one NO2 group can be used as a substrate for detecting nitroreductase enzyme activity in a composition and allows for the use of a nitroreductase enzyme in an enzyme-reporter system for the detection of analytes, binding reactions or gene expression.

L'invention concerne un procédé servant à accroître la fluorescence d'une molécule colorante de cyanine contenant au moins un groupe NO2, caractérisé par la réduction d'au moins un groupe NO2 en NHOH ou en NH2 au moyen d'une nitroréductase. La molécule colorante de cyanine comporte au moins un groupe NO2 pouvant être utilisé comme substrat pour détecter l'activité enzymatique d'une nitroréductase dans une composition, et permet d'utiliser une nitroréductase dans un système de rapporteur enzymatique pour la détection d'analytes, dans des réactions de liaison ou dans l'expression génique.
