A follistatin mutant which can specifically inhibit the activity of GDF-8 without inhibiting the activity of activin. The follistatin mutant polypeptide is characterized by (a) containing follistatin domain I, (b) having an amino acid sequence represented by the formula (I): Cys-(X1)a-Cys-(X2)b-Cys-(X3)c- Cys-(X4)d-Cys-(X5)e-Cys-(X6)f-Cys-(X7)g-Cys-(X8)h-Cys-(X9)i-Cys (wherein X1, X2, X3, X4, X5, X6, X7, X8, and X9 are the same or different and each independently represents a residue of an amino acid present in the nature, other than cysteine; a is an integer of 2-6; b is an integer of 3-7; c is an integer of 7-11; d is an integer of 0-4; e is an integer of 1-6; f and g each is an integer of 8-12; h is an integer of 4-8; and i is an integer of 11-15) and containing no amino acid sequence having 50% or higher homology to the amino acid sequence of sequence number 2 or 29, and (c) inhibiting the activity of GDF-8 more selectively than the activity of activin.

Le polypeptide mutant de follistatine est caractérisé (a) en ce qu'il contient une follistatine de domaine I, (b) en ce qu'il possède une séquence d'acides aminés représentée par la formule (I) : Cys-(X<1>)a-Cys-(X<2>)b-Cys-(X<3>)c-Cys-(X<4>)d-Cys-(X<5>)e-Cys-(X<6>)f-Cys-(X<7>)g-Cys-(X<8>)h-Cys-(X<9>)i-Cys (où X<1>, X<2>, X<3>, X<4>, X<5>, X<6>, X<7>, X<8>, et X<9> sont identiques ou différents et représentent chacun indépendamment un résidu d'acide aminé présent dans la nature, autre que la cystéine ; a représente un entier de 2 à 6 ; b représente un entier de 3 à 7 ; c représente un entier de 7 à 11 ; d représente un entier de 0 à 4 ; e représente un entier de 1 à 6 ; f et g sont chacun un entier de 8 à 12 ; h représente un entier de 4 à 8 ; et i est un entier de 11 à 15) et ne contenant aucune séquence d'acides aminés d'une homologie supérieure ou égale à 50 % par rapport à la séquence d'acides aminés d'un numéro de séquence 2 ou 29, et (c) en ce qu'il inhibe l'activité de GDF-8 de manière plus sélective que l'activité de l'activine.
