Variants of hedgehog protein that contain N-terminal modifications are described that can block hedgehog function; thus allowing these variants to serve as functional antagonists. These peptides have a primary amino acid sequence lacking the ability to elicit a hedgehog-dependent response in C3H 10T1/2 cells but having the ability to bind to the hedgehog receptor, patched-1. Methods for producing such functional antagonists and methods of using the funtional antagonists are also described.

L'invention concerne des variants de la protéine hedgehog comportant des modifications N-terminales, capables de bloquer la fonction hedgehog, ce qui leur permet ainsi de jouer un rôle d'antagonistes fonctionnels. Ces peptides comportent une séquence primaire d'acides aminés ne pouvant pas éliciter une réponse dépendant du hedgehog dans les cellules C3H 10T1/2, mais pouvant néanmoins se lier au récepteur hedgehog, patched-1. Sont également décrites des techniques de production de ces antagonistes fonctionnels et des techniques concernant leur utilisation.
