ABSTRACT OF THE DISCLOSURE Methods are provided for obtaining, from fungi selected from the dematiaceous hyphomycetes, chloroperoxidases, which can catalyze oxidation of chloride, bromide and iodide in aqueous solution in the presence of H202 and which have optimum activity above about pH 5. Further, methods are provided for obtaining from microorganisms, including dematiaceous hyphomycetes, non-heme-containing chloroperoxidases, which have the same halide-oxidation capabilities and pH optima and which are substantially more stable to inactivation by H202 and HOCl than the chloroperoxidase of Caldariomyces fumago. The chloroperoxidases obtained by the methods of the invention are used, in aqueous solution at a pH between about 4 and 9 in the presence of H202, to catalyze the oxidation of iodide to iodine and the same halogenation reactions, between halide ions and organic acceptor compounds, that can be catalyzed by C. fumago chloroperoxidase. The chloroperoxidases can be used in purified form or as part of undisrupted cells.
