The present invention relates to a plant mangrove-associated fungus Culvularia lunata of International deposition no. MTCC 5109, and a simple and efficient method of obtaining high yield of pure mannitol from a plant mangrove- associated fungus Curvularia lunata, said method comprising steps of cutting the leaves into small pieces and placing it on potato dextrose agar (PDA) plates for 48 hours, maintaining the culture at temperature ranging between 26- 470C and salinity ranging between 4-32 ppt with occasional stress conditions for about 17-19 days to obtain mycelial mat, sonicating the mat to lyse the cells, extracting crude from the sonicated mat using methanol repeatedly, concentrating the crude, treating the concentrated crude extract with solvents of increasing polarity, obtaining aqueous fraction as white powder residue after the treatment containing crude mannitol, purifying the crude mannitol by chromatography to obtain pure mannitol with about 75% of the total crude extract.

Selon ce procédé, on découpe les feuilles en petits morceaux et on les place sur des plaques de gélose dextrosée à la pomme de terre pendant 48 heures, on maintient la culture à une température comprise entre 26-47 ·C et à une salinité comprise entre 4-32 ppt, sous des conditions occasionnelles de stress pendant 17-19 jours, jusqu'à produire une trame mycélienne, on soumet la trame à un traitement aux ultrasons afin de lyser les cellules, on extrait le brut de la trame traitée aux ultrasons en utilisant du méthanol à plusieurs reprises, on concentre le brut, on traite l'extrait brut concentré avec des solvants pour accroître la polarité, on produit une fraction aqueuse sous forme d'une poudre blanche résiduelle après le traitement, la poudre blanche résiduelle contenant du mannitol brut, et on purifie le mannitol brut par chromatographie jusqu'à obtenir du mannitol pur avec environ 75 % de l'extrait brut total.
