The invention concerns fused heterocyclic isoxazoline derivatives of Formula (I), the pharmaceutically acceptable acid or base addition salts thereof, the stereochemically isomeric forms thereof and the N-oxide form thereof, more in particular, tetrahydropyranoisoxazole, hexahydroisoxazolopyridine, tetrahydrothiopyrano isoxazole and hexahydrobenzoisoxazole derivatives fused to a heterocyclic ring system via the 6-membered ring of the bicyclic moiety as well as processes for their preparation, pharmaceutical compositions comprising them and their use as a medicine, in particular for treating depression, anxiety, movement disorders, psychosis, Parkinson's disease and body weight disorders including anorexia nervosa and bulimia, wherein the variables are defined as in Claim 1. The compounds have surprisingly been shown to have selective serotonine (5-HT) reuptake inhibitor activity as well as .alpha.2-adrenoceptor antagonist activity, compounds according to the invention are also suitable for treatment and/or prophylaxis in diseases where either one of the activities alone or the combination of said activities may be of therapeutic use.
L'invention concerne des dérivés d'isoxazoline hétérocycliques représentés par la formule (I), leurs sels d'addition d'acide ou de base pharmaceutiquement acceptables, leurs formes stéréochimiquement isomères et leur forme N-oxyde. Plus particulièrement, elle se rapporte à des dérivés de tetrahydropyranoisoxazole, hexahydroisoxazolopyridine, tetrahydrothiopyrano isoxazole et de hexahydrobenzoisoxazole fusionnés avec un système de noyau hétérocyclique au moyen du noyau à 6 chaînons de la fraction bicyclique, ainsi que sur des procédés de préparation de compositions pharmaceutiques les contenant, et sur leur utilisation en tant que médicaments, notamment dans le traitement de la dépression, de l'anxiété, des troubles des mouvements, de la psychose, de la maladie de Parkinson et de troubles alimentaires tels que l'anorexie nerveuse, la boulimie, ces variables étant définies dans la revendication 1. Ces composés ont étonnement prouvé posséder une activité inhibitrice de recaptage de sérotonine (5-HT), ainsi qu'une activité antagoniste du ?¿2?-adréno-récepteur, les composés selon l'invention étant aussi utiles dans le traitement et/ou la prophylaxie de maladies pour lesquelles une seule des activités ou la combinaison de ces activités peut présenter une utilité thérapeutique.
