Targeted transcriptional effectors (tran-scription activators and transcription repressors) derived from meganucleases are described. Also described are nu-cleic acids encoding same, and methods of using same to regulate gene expression. The targeted transcriptional ef-fectors can comprise (i) a meganuclease DNA-binding do-main lacking endonuclease cleavage activity that binds to a target recognition site; and (ii) a transcription effector do-main.

L'invention porte sur des effecteurs de transcription ciblés (activateurs et répresseurs de transcription) issus de méganucléases. L'invention porte également sur des acides nucléiques codant pour ceux-ci et sur des procédés d'utilisation de ceux-ci pour réguler l'expression génique. Les effecteurs de transcription ciblés peuvent comporter (i) un domaine de liaison à l'ADN de méganucléase n'ayant pas d'activité de clivage d'endonucléase qui se lie à un site de reconnaissance cible, et (ii) un domaine effecteur de transcription.
