The present invention relates to granulocyte colony stimulating factor ("G- CSF") analog polypeptide compositions, related nucleic acids, expression constructs, host cells, and processes for recombinant production of the present G-CSF analogs. The concept detailed herein involves novel mutants of G- CSF, using single substitutions to amino acids, which were rationally chosen to affect the cellular trafficking of G-CSF and/or G-CSFR. In addition, pharmaceutical compositions, and methods of use are provided.

La présente invention se rapporte à des compositions polypeptidiques d'analogues du facteur stimulant les colonies de granulocytes ("G-CSF"), à des acides nucléiques associés, à des produits recombinés d'expression, à des cellules hôtes et à des procédés permettant la production par recombinaison des présents analogues du G-CSF. L'invention se rapporte également à de nouveaux mutants du G-CSF ne comportant que des substitutions simples d'acides aminés, qui ont été sélectionnées rationnellement de manière à modifier le trafic cellulaire du G-CSF et/ou du G-CSFR. L'invention se rapporte en outre à des compositions pharmaceutiques et à des procédés d'utilisation de telles compositions pharmaceutiques.
