Pharmaceutical gastroretentive drug delivery system for the controlled release of an active agent in the gastrointestinal tract, which comprises: (a) a single- or multi-layered matrix having a two- or three-dimensional geometric configuration comprising a polymer that does not retain in the stomach more than a conventional dosage form selected from (1) degradable polymers that may be hydrophilic polymers not instantly soluble in gastric fluids, enteric polymers substantially insoluble at pH less than 5.5 and/or hydrophobic polymers and mixtures thereof; (2) non-degradable polymers; and any mixtures of (1) and (2); (b) a continuous or non-continuous membrane, that does not retain in the stomach more than a conventional dosage form, affixed or attached to said matrix, the membrane comprising at least one polymer having a substantial mechanical strength; and (c) a drug; wherein the matrix when affixed or attached to said membrane prevents evacuation from the stomach of said delivery system for a period of time of from about 3 to about 24 hours. The system may further comprise a shielding layer. The delivery system is particularly suitable for the delivery of drugs having a narrow absorption window in the gastrointestinal tract, drugs intended for local treatment of the gastrointestinal tract and drugs degraded in the colon, and is intended for human and veterinary use.
L'invention porte sur un système pharmaceutique d'administration de médicament contre la rétention gastrique, système s'appliquant à un agent actif à libération régulée dans le tube digestif. Ce système comprend : (a) une matrice à couche unique ou à plusieurs couches dont la configuration géométrique bi- ou tridimensionnelle comprend un polymère qui ne retient pas dans l'estomac plus d'une forme posologique traditionnelle sélectionnée parmi (1) des polymères dégradables qui peuvent être des polymères hydrophiles non solubles instantanément dans des fluides gastriques, des polymères entériques pratiquement insolubles et ayant un pH inférieur à 5,5 et/ou des polymères hydrophobes et des mélanges de ceux-ci ; (2) des polymères non dégradables ; et des mélanges quelconques de (1) et (2) ; (b) une membrane continue ou non continue qui ne retient pas dans l'estomac plus d'une forme posologique traditionnelle, collée ou attachée à la matrice, cette membrane comprenant au moins un polymère ayant une grande résistance mécanique ; et (c) un médicament. La matrice, collée ou attachée à la membrane, empêche le système d'administration de s'évacuer de l'estomac entre environ 3 et environ 24 heures. Le système peut également comprendre une couche de protection. Le système d'administration est notamment approprié pour administrer des médicaments présentant une fenêtre d'absorption étroite dans le tube digestif, des médicaments destinés à traiter localement le tube digestif et des médicaments se dégradant dans le côlon. Ce système peut être destiné à un usage humain et vétérinaire.
